2015年3月24日、FDAは混合ジフテリア・破傷風トキソイド・無細胞百日咳吸着(DTaP)・不活化ポリオウイルス(IPV)ワクチン(DTaP-IPV、Quadracel)を認可した。Quadracelは、4～6歳の小児に対する使用をアメリカで承認された2番目のDTaP-IPVである。また、DTaP-IPV-Hib(Pentacel)やDTaP(Daptacel)の投与を4回受けた4～6歳児に対し、 DTaPシリーズの5回目またはIPVシリーズの4回または5回目の投与として承認されている。Quadracel投与量中に含まれる抗原[ジフテリア、破傷風トキソイド、百日咳抗原(百日咳トキソイド、線維状赤血球凝集素、パータクチン、線毛2型または3型)、不活化ポリオウイルス1～3型]は、DTaP-IPV-Hib(Pentacel)と一致している。Quadracelは防腐剤を含まず、筋肉注射により投与される。アメリカの4～6歳の小児において実施された臨床試験では、Quadracelと別々に投与されたDTaP(Daptacel)およびIPV(Ipol)ワクチンの安全性および反応原プロファイルは、麻疹、おたふく風邪、風疹、水痘ワクチンの同時投与された2回目の接種の有無にかかわらず同等であった。Quadracelは4歳未満および7歳以上の小児に投与されるべきではない。もし、何らかの事情によりDTaPやIPVの早期の投与のために4歳未満で投与される時、および最小の投与間隔が必要とされる時は、その投与はDTaPやIPVシリーズの有効な回数としてみなされ、追加の投与は必要ないが、IPVシリーズの最終投与は過去の投与回数にかかわらず4歳以上で投与し、前回からの投与間隔は最低6カ月必要である。異なる製造者からのDTaPワクチンの交換の安全性および免疫原生に関するデータは限られている。ACIPは、可能であれば同じ製造者のDTaP ワクチンをシリーズ中の各回の投与に使用することを推奨している。
